Why model Cd14
Cd14 encodes the mouse ortholog of human CD14, a leucine-rich repeat horseshoe glycoprotein present both as a GPI-anchored surface receptor on monocytes, macrophages, and neutrophils and as a soluble plasma form generated by shedding or direct secretion. Its function is ligand delivery rather than signaling: CD14 accepts lipopolysaccharide extracted from bacterial membranes by LBP and loads lipid A into the TLR4 and MD-2 (Ly96) complex, enabling both MyD88 dependent surface responses and CD14 dependent endocytosis that licenses TRIF and type I interferon output, and it similarly assists TLR2 lipopeptide recognition and nucleic acid sensing TLR trafficking. Nulls are viable and profoundly hyporesponsive to low-dose LPS while retaining responses to high-dose stimulation, a dose-dependent phenotype that must be built into endotoxemia study design. Humanization is correct for anti-CD14 antibody and sepsis therapeutic programs, whereas conditional deletion with myeloid or hepatocyte Cre partitions membrane and soluble CD14 contributions.
