Why model Chd4
Chromodomain helicase DNA-binding protein 4, encoded by Chd4 and orthologous to human CHD4, supplies the Mi-2 beta ATPase motor of the nucleosome remodeling and deacetylase complex, partnering with HDAC1 and HDAC2, MTA1 through MTA3, MBD2 or MBD3, GATAD2, and RBBP4. Tandem PHD fingers read the H3 tail while paired chromodomains grip nucleosomal DNA, coupling remodeling to deacetylation for enhancer decommissioning and promoter silencing, and enabling rapid PARP-dependent recruitment to double-strand breaks. Recurrent missense mutations in the ATPase and chromodomain regions occur in endometrial and serous carcinoma, indicating separation of function rather than simple loss. Constitutive deletion is early embryonic lethal, so conditional alleles with tissue-restricted Cre are required, and ATPase-dead knockins distinguish catalytic from scaffolding roles. Chd3 and Chd5 form mutually exclusive NuRD variants and partially buffer loss, so consider compound alleles when a single deletion yields a muted chromatin phenotype.
