Cntnap2, ortholog of human CNTNAP2, encodes CASPR2, a neurexin superfamily single-pass adhesion molecule whose ectodomain combines discoidin, laminin G, EGF and fibrinogen-like modules and whose short cytoplasmic tail binds protein 4.1B; at myelinated axon juxtaparanodes it forms a ternary complex with TAG-1 (Contactin-2) to cluster Kv1.1 and Kv1.2 channels and prevent aberrant reexcitation, and it also functions at cortical synapses and in interneuron migration. Recessive truncating alleles cause cortical dysplasia focal epilepsy syndrome, common variants associate with language and autism phenotypes, and the CASPR2 ectodomain is a major autoantigen in limbic encephalitis and Morvan syndrome. Constitutive nulls are viable and reproduce interneuron loss, abnormal ultrasonic vocalization and repetitive behavior, giving a workable model, while a humanized ectodomain allele uniquely supports patient autoantibody and immunotherapy studies. Note the extreme locus size, over two megabases, which constrains full-locus humanization and favors targeted domain replacement with careful conditional exon design.
