Deptor, mouse ortholog of human DEPTOR, encodes a naturally occurring inhibitor of both mTOR complexes, using tandem DEP domains and a C-terminal PDZ domain that binds the FAT domain of mTOR to suppress mTORC1 and mTORC2 catalytic output toward S6K1, 4E-BP1 and AKT serine 473. Regulation is largely post-translational: active mTORC1 and CK1 alpha phosphorylate DEPTOR to create a beta-TrCP degron, so growth factor stimulation degrades its own brake in a feedforward loop, while nutrient withdrawal restores it. Loss of DEPTOR relieves mTORC1 inhibition but also removes the negative feedback that normally suppresses PI3K and AKT, so overexpression in a subset of multiple myeloma paradoxically sustains AKT activity, a genuinely counterintuitive result that allele design must respect. Constitutive nulls are viable, permitting simple alleles, but conditional deletion or degron-resistant knockins are far more informative for dissecting feedback. Match allele to endpoint, using knockins when the question is loop architecture rather than net mTOR flux.
