Why model Elane
Elane is the mouse ortholog of human ELANE and encodes neutrophil elastase, a chymotrypsin-fold serine protease packaged into azurophilic granules after cathepsin C dependent propeptide removal, where it degrades elastin, collagens, and bacterial outer membrane protein A, inactivates tissue factor pathway inhibitor, processes chemokines, and drives chromatin decondensation during neutrophil extracellular trap formation, restrained extracellularly by alpha-1 antitrypsin and SLPI. The disease logic here is misfolding rather than lost proteolysis: human ELANE mutations cause severe congenital and cyclic neutropenia through mistrafficking and unfolded protein response driven apoptosis of promyelocytes, so an Elane null does not phenocopy the human condition and knockin of specific patient alleles, ideally within a humanized locus, is the only faithful design. Cathepsin G and proteinase 3 overlap for extracellular substrate degradation, meaning compound alleles are needed for protease-driven tissue injury studies, while sivelestat class inhibitor programs require humanization for potency assessment.
