Why model Epas1

Epas1, the mouse ortholog of human EPAS1, encodes HIF2 alpha, a bHLH-PAS transcription factor that dimerizes with ARNT and, in normoxia, is hydroxylated at prolines 405 and 531 by EGLN1 and related prolyl hydroxylases, creating the VHL recognition site for proteasomal destruction, while FIH hydroxylation of an asparagine blocks p300 recruitment. HIF2 alpha drives a target set biased toward erythropoietin, VEGFA and the endothelial and renal programs, distinct from the glycolytic bias of HIF1 alpha. Gain of function mutations at the hydroxylation sites cause erythrocytosis with paraganglioma and pheochromocytoma, and the druggable PAS-B cavity is the basis of belzutifan, so a knockin of a hydroxylation-resistant allele such as the human P531 variant is the appropriate tool for testing HIF2 alpha selective inhibitors. Null mice show strain-dependent embryonic and perinatal lethality from catecholamine and vascular defects, so conditional deletion is preferable. Hif1a occupies overlapping HREs, making double conditional alleles necessary for isoform assignment.
