F2, ortholog of human F2, encodes prothrombin, the vitamin K dependent precursor of thrombin and the central protease of hemostasis. Prothrombinase, comprising factor Xa and factor Va on an anionic phospholipid surface, cleaves prothrombin at two sites to release the Gla and kringle containing fragment 1.2 and generate alpha-thrombin, whose substrate selection is governed by two electropositive exosites: exosite I engages fibrinogen, PAR1, and thrombomodulin, while exosite II binds heparin and GPIb alpha. Thrombin converts fibrinogen to fibrin, activates factors V, VIII, XI, and XIII along with platelet PAR1 and PAR4, and when bound to thrombomodulin switches specificity to activate protein C. Complete deficiency is lethal in mouse, with embryonic loss and neonatal hemorrhage, so hypomorphic or conditional alleles are mandatory. Note that the common human G20210A thrombophilia allele lies in the 3 prime untranslated region and raises expression rather than altering sequence, so dosage-increasing designs, not missense knockins, are the faithful mimic.
