Gdf11 Conditional Knockout mouse models

Gdf11, the mouse ortholog of human GDF11, encodes a TGF-beta superfamily ligand secreted as a proprotein whose furin-cleaved mature dimer remains latent in a noncovalent complex with its own prodomain until BMP1 and tolloid proteases liberate it. Active GDF11 engages ActRIIA and ActRIIB with ALK4 or ALK5 to phosphorylate SMAD2 and SMAD3, and its mature domain is roughly 90 percent identical to myostatin, so the two ligands share receptors and the antagonist follistatin. Loss of Gdf11 shifts anterior Hox boundaries, producing extra thoracic vertebrae, kidney agenesis, and perinatal lethality, which makes conditional nulls rather than germline deletion the practical route to adult physiology. Prodomain or cleavage-site knockins interrogate latency and proteolytic activation separately from total ligand loss, while humanizing the mature domain supports selectivity testing of antibodies meant to spare myostatin. Because Mstn compensates functionally, pair the allele with a tissue-restricted Cre and consider compound Mstn genetics before interpreting muscle endpoints.

Tissue restricted knockout of Gdf11 keeps wild type function everywhere else. Breeding is often more robust, and the setup mirrors patient biology where mutations arise in a subset of cells.
