Prodomain or cleavage-site knockins interrogate latency and proteolytic activation separately from total ligand loss, while humanizing the mature domain supports selectivity testing of antibodies meant to spare myostatin. Because Mstn compensates functionally, pair the allele with a tissue-restricted Cre and consider compound Mstn genetics before interpreting muscle endpoints.
