Why model Grb2
Grb2, the mouse ortholog of human GRB2, encodes a compact adaptor built from a central SH2 domain flanked by two SH3 domains, with no catalytic activity of its own; the SH2 domain reads phosphotyrosine in pYXNX motifs on activated receptors such as Egfr and on Shc, while the SH3 domains bind proline-rich sequences in Sos1 and Gab1, thereby converting receptor autophosphorylation into Ras nucleotide exchange and, through Gab1, into PI3K activation. Homozygous deletion causes early embryonic lethality around gastrulation, so conditional alleles are required for any postnatal or tissue-specific study, and separation-of-function knockins are particularly informative here: an SH2-binding-dead substitution uncouples receptor docking, while selective disruption of the Sos1-binding or Gab1-binding SH3 surfaces splits Erk from Akt output within the same receptor. The related adaptors Grap and Grap2 overlap in hematopoietic lineages and can mask loss, so consider compound designs and a driver validated in the lineage under study.
