Grn, the mouse ortholog of human GRN, encodes progranulin, a secreted cysteine-rich glycoprotein composed of seven and a half granulin repeats that is delivered to lysosomes through sortilin (Sort1) and prosaposin, then proteolytically processed by cathepsins and elastase into individual granulin peptides with activities distinct from the intact precursor. Human genetics defines a dosage logic that should drive allele choice: heterozygous nonsense and frameshift mutations cause frontotemporal dementia with TDP-43 pathology through haploinsufficiency, whereas complete biallelic loss causes neuronal ceroid lipofuscinosis type 11. Null mice are viable and accumulate lipofuscin with age-dependent microgliosis, so germline heterozygotes test haploinsufficiency, full nulls test lysosomal storage, and humanized knockins support progranulin-raising biologics and sortilin-blocking antibodies. No paralog compensates, making dose titration rather than redundancy the central experimental variable.
