H3f3a, ortholog of human H3F3A, encodes histone variant H3.3, which differs from replication-coupled H3.1 at serine 31 and across the alanine 87, isoleucine 89, glycine 90 patch that dictates chaperone selection, permitting HIRA-mediated deposition at transcribed gene bodies and regulatory elements and ATRX with DAXX deposition at telomeres and heterochromatin outside S phase. Its oncohistone alleles are dominant and lesion specific: lysine 27 to methionine sequesters and inhibits the EZH2 active site, collapsing global H3K27 trimethylation in diffuse midline glioma, while glycine 34 to arginine or valine acts in cis by impairing SETD2 and KDM4 activity on the same tail in hemispheric pediatric glioblastoma. Use conditional Lox-Stop-Lox knockin of the specific substitution at the endogenous locus with a spatially restricted Cre, since one mutant copy among many histone genes suffices. The identical protein from H3f3b buffers null alleles, so loss-of-function work requires compound targeting.
