Kcnj12, the mouse ortholog of human KCNJ12, encodes Kir2.2, a strong inward rectifier whose rectification arises from voltage-dependent block of the pore by intracellular magnesium and polyamines binding at a conserved aspartate in the second transmembrane segment and at cytoplasmic domain residues. Kir2.2 requires PIP2 for opening, and the crystal structures of this channel defined the PIP2 binding site for the entire Kir2 family. Functionally it co-assembles with Kir2.1 and Kir2.3 to build the cardiac and neuronal IK1 conductance that stabilizes resting membrane potential and sets the final phase of repolarization, and it is expressed in ventricular myocytes, skeletal muscle, and brain. Because heteromerization with the Kcnj2 and Kcnj4 products is the rule rather than the exception, single-locus nulls are typically viable with modest current reduction, and dominant-negative pore knockins or compound conditional alleles are needed to collapse IK1. Choose conditional deletion with cardiac or neuronal Cre when attributing arrhythmia or excitability phenotypes.
