Why model Kcnj3

Encoding GIRK1 (Kir3.1), Kcnj3 is the mouse ortholog of human KCNJ3 and supplies a subunit that cannot form functional homotetramers and instead obligately partners with GIRK2 or GIRK4 to build G protein-gated inwardly rectifying potassium channels. These channels are opened directly by Gbeta gamma dimers released from Gi and Go-coupled receptors, including GABA-B, M2 muscarinic, opioid, adenosine A1, and D2 receptors, with PIP2 and intracellular sodium acting as required cofactors and RGS proteins setting deactivation kinetics. GIRK1-containing channels mediate slow inhibitory postsynaptic currents in hippocampus and cortex and the atrial IKACh that slows heart rate. Because the subunit is obligatorily heteromeric, deletion removes surface channels rather than reducing a parallel conductance, and conditional alleles with forebrain or interneuron Cre drivers cleanly separate central from cardiac phenotypes. Kcnj6 and Kcnj5 products remain partially functional without GIRK1, so quantify residual Gbeta gamma-activated current, and use humanized alleles for GIRK-selective modulator campaigns.
