Why model Kcnq3
Kcnq3, the mouse ortholog of human KCNQ3, encodes Kv7.3, the obligatory heteromeric partner that increases M current amplitude tenfold when co-assembled with Kv7.2 or Kv7.5, largely because its carboxy-terminal coiled-coil stabilizes tetramer assembly and its higher PIP2 affinity keeps the channel available at physiological lipid levels. Kv7.3 also carries the alanine at position 315 in the pore that is critical for potassium selectivity and for the action of openers such as retigabine, which binds a conserved tryptophan in S5. Like Kv7.2, the channel is concentrated at the axon initial segment and nodes through ankyrin-G binding, where it sets spike threshold and adaptation.
