Distinct germline missense variants cause X-linked intellectual disability syndromes including the Opitz-Kaveggia and Lujan phenotypes. Because germline deletion is early embryonic lethal, conditional nulls address transcriptional dependency in defined lineages while precise hotspot knockins are required for leiomyoma modeling and CDK8 inhibitor testing. The paralog Med12l overlaps partially in neural tissue, and X-linkage means sex and Cre selection (for example Amhr2-cre for myometrium) must be predefined.
