Why model Mtor

Mtor, the mouse ortholog of human MTOR, encodes a phosphatidylinositol kinase-related serine and threonine kinase with HEAT repeats, FAT, FRB, kinase, and FATC domains that nucleates two distinct complexes: mTORC1, containing Raptor and Mlst8, is recruited to lysosomes by Rag GTPases in response to amino acids and activated by Rheb-GTP under growth factor and energy input, phosphorylating Rps6kb1 and Eif4ebp1 to drive translation while suppressing Ulk1-dependent autophagy; mTORC2, containing Rictor, Mapkap1, and Mlst8, phosphorylates Akt at Ser473. Rapamycin acts allosterically by bridging Fkbp12 to the FRB domain, inhibiting mTORC1 acutely and mTORC2 only chronically. Homozygous deletion is lethal shortly after implantation, so conditional alleles are required, and the rapamycin-resistant Ser2035Thr knockin remains the definitive genetic test of on-target drug action. Kinase-dead and activating knockins separate scaffolding from catalysis, while Rptor and Rictor floxed alleles resolve complex-specific questions that Mtor deletion cannot.
