Pdk3 Conditional Knockout mouse models

Pdk3, the mouse ortholog of human PDK3, encodes the pyruvate dehydrogenase kinase isoform with the highest intrinsic specific activity toward E1 alpha and the strongest dependence on binding the lipoyl-bearing L2 domain of the E2 core for full activation, a property that renders it comparatively resistant to feedback inhibition by pyruvate. That biochemistry carries clinical weight, since the gene is X-linked and gain-of-function missense variants strengthening L2 binding cause X-linked Charcot-Marie-Tooth neuropathy type 6 through chronic PDH hyperphosphorylation and suppressed oxidative flux. The design fork is therefore unusual for this family: a hyperactivating patient-matched knockin, not a null, reproduces the human disease, while a conditional null answers the separate question of where this isoform normally restrains PDH. Because Pdk1, Pdk2 and Pdk4 overlap substantially, a single knockout is easily buffered and compound alleles should be planned. Match allele to endpoint, pairing peripheral nerve analysis with the knockin and flux measurements with the null.

Pdk3 conditional knockout mice carry a floxed allele so you delete function only where Cre is active. The germline allele stays intact until you cross to a tissue specific Cre.
