Pgk1 is the mouse ortholog of human PGK1 and encodes phosphoglycerate kinase 1, the enzyme that transfers the high-energy phosphate of 1,3-bisphosphoglycerate to ADP, generating the first net ATP of glycolysis and yielding 3-phosphoglycerate that also feeds the serine biosynthesis branch. The gene is X-linked and ubiquitously expressed, and the protein moonlights beyond canonical glycolysis, acting as a disulfide reductase that liberates angiostatin from plasmin and, in some settings, entering mitochondria to phosphorylate pyruvate dehydrogenase kinase 1. Recessive human deficiency produces hemolytic anemia, myopathy and neurological disease with hemizygous male presentation. Because complete loss removes a non-redundant ATP-generating step, conditional floxed alleles are the practical format, with patient-matched knockins preserving residual activity for erythroid and muscle phenotyping.
