Serpind1 is the mouse ortholog of human SERPIND1 and encodes heparin cofactor II, a plasma serpin that inhibits thrombin through a Leu-Ser reactive center and is accelerated up to a thousand fold by dermatan sulfate, which binds a basic surface and releases an acidic N-terminal extension that then engages thrombin anion-binding exosite II. This dermatan sulfate dependence localizes activity to the subendothelial matrix rather than the luminal heparan sulfate compartment served by antithrombin, giving heparin cofactor II a distinct role after vessel injury. Nulls are viable and fertile with normal baseline hemostasis but show accelerated thrombus formation after photochemical or ferric chloride carotid injury, so germline deletion is informative while hepatocyte conditional alleles pin the source. Glycosaminoglycan-binding or reactive center loop knockins separate cofactor dependence from intrinsic inhibition. Since Serpinc1 dominates fluid-phase thrombin control, interpret accordingly and align alleles with arterial injury or restenosis endpoints.
