Slc1a3 knockout mice are viable but show cerebellar motor incoordination, heightened susceptibility to excitotoxic injury, and retinal dysfunction. Allele choice therefore forks between conditional null for regional uptake capacity questions and patient knockins for the dominant channelopathy mechanism. Because Slc1a2 carries most forebrain uptake, cerebellar and retinal endpoints are the most informative, and inducible glial CreERT2 drivers avoid developmental confounds.
