Why model Stat1
Stat1 encodes the ortholog of human STAT1, a latent cytoplasmic transcription factor recruited through its SH2 domain to phosphotyrosine docking sites on activated cytokine receptors, phosphorylated at Tyr701 by JAK1 and JAK2, and then assembled either as the homodimeric GAF complex that binds GAS elements downstream of interferon gamma or, with STAT2 and IRF9, as ISGF3 which binds interferon stimulated response elements downstream of type I interferon. Ser727 phosphorylation in the transactivation domain provides a second, JAK-independent input required for full activity. Human loss-of-function alleles cause mycobacterial and viral susceptibility while gain-of-function alleles cause chronic mucocutaneous candidiasis, so knockins of specific coiled-coil or DNA-binding domain substitutions answer questions deletion cannot. Nulls are viable but profoundly infection susceptible and tumor prone, making constitutive alleles usable, with conditional nulls reserved for separating hematopoietic from stromal interferon responses in tumor immunology.
