Ser727 phosphorylation in the transactivation domain provides a second, JAK-independent input required for full activity. Human loss-of-function alleles cause mycobacterial and viral susceptibility while gain-of-function alleles cause chronic mucocutaneous candidiasis, so knockins of specific coiled-coil or DNA-binding domain substitutions answer questions deletion cannot. Nulls are viable but profoundly infection susceptible and tumor prone, making constitutive alleles usable, with conditional nulls reserved for separating hematopoietic from stromal interferon responses in tumor immunology.
