Why model Stat5a
Stat5a is the mouse ortholog of human STAT5A and lies in a tandem duplication with Stat5b, an arrangement that dictates targeting strategy since the two genes are separated by only a few kilobases and share more than ninety percent identity. The protein is recruited by its SH2 domain to phosphotyrosines on prolactin, growth hormone, erythropoietin and common gamma chain cytokine receptors, phosphorylated by JAK2 or JAK1 at a carboxy-terminal tyrosine, then dimerizes or tetramerizes on GAS elements to drive Cish, Bcl2l1 and casein gene transcription. Selective loss produces failed mammary lobuloalveolar development with largely preserved body growth, the reciprocal of Stat5b deletion. Because paralog compensation is substantial, rigorous work generally uses floxed alleles spanning both genes with lineage-restricted Cre drivers, or tetramerization-deficient amino-terminal domain knockins when the question is target gene selectivity rather than total signaling capacity.
