Why model Ang
Angiogenin, the product of mouse Ang and ortholog of human ANG (also called RNASE5), is a secreted RNase A superfamily ribonuclease whose His13, Lys40, His114 catalytic triad supports weak but obligatory cleavage of tRNA anticodon loops, generating stress-induced tiRNA fragments that displace eIF4G and reprogram translation initiation. Under basal conditions the enzyme is clamped by cytosolic RNH1, and during stress it is internalized by motor neurons through syndecan and neuropilin-1 before translocating to the nucleolus to sustain 45S rRNA transcription. Amyotrophic lateral sclerosis and frontotemporal dementia alleles such as K40I and C39W are catalytically hypomorphic, so a conditional null addresses cell-autonomous requirement while a patient point knockin tests whether diminished ribonucleolytic output alone drives motor neuron vulnerability, and humanization enables recombinant protein dosing work. Because the mouse locus is a tandem cluster spanning Ang2 through Ang6, single-gene deletion is buffered; pair Chat-Cre or Gfap-Cre conditionals with neuromuscular junction and survival endpoints.
