Why model Arnt
Arnt, the mouse ortholog of human ARNT, encodes HIF1 beta, the obligate class II bHLH-PAS dimerization partner that licenses DNA binding by HIF1 alpha, EPAS1 and the aryl hydrocarbon receptor, contacting hypoxia response elements or xenobiotic response elements only as a heterodimer and recruiting p300 and CBP through partner transactivation domains. Because ARNT is constitutively expressed and not oxygen regulated, it functions as the shared limiting hub where hypoxic and xenobiotic transcriptional programs converge and compete. Germline deletion is embryonic lethal near midgestation from failed placental vascularization and yolk sac angiogenesis, so conditional alleles are essential for adult work in hepatocytes, pancreatic beta cells, keratinocytes and hematopoietic compartments. Because Arnt2 substitutes in neurons and some endocrine tissues, CNS phenotypes require compound conditional genetics to avoid false negatives. Deleting Arnt collapses the entire hypoxic and AHR axis simultaneously, so when isoform-specific questions matter, target Hif1a or Epas1 directly instead of this shared partner.
