Atp5md corresponds to human ATP5MD, also catalogued as USMG5 and long described as DAPIT, the diabetes associated protein in insulin sensitive tissue, a small hydrophobic subunit of the membrane embedded Fo sector of mitochondrial ATP synthase. DAPIT associates with the c-ring periphery and is required for efficient accumulation of assembled complex V, and its depletion lowers holoenzyme content and oligomycin sensitive ATP synthesis without eliminating individual core subunits, indicating a role in stabilizing and oligomerizing the enzyme rather than in catalysis itself. Human loss of function through a splice altering variant causes Leigh syndrome with isolated complex V deficiency. Design accordingly: a conditional null tests whether a tissue tolerates reduced holoenzyme, while a splice site or missense knockin reproduces graded human deficiency. Because expression is highest in muscle, heart, and adipose, pair Ckmm-Cre, Myh6-MerCreMer, or Adipoq-Cre with respirometry, cristae ultrastructure, and glucose tolerance endpoints.
