Bak1 encodes BAK, the mouse ortholog of human BAK1 and one of two obligate multidomain effectors of mitochondrial outer membrane permeabilization. Unlike BAX, BAK resides constitutively in the outer membrane anchored by its alpha9 transmembrane helix, and activation begins when a BH3-only ligand such as BID, BIM, or PUMA transiently engages the canonical hydrophobic groove, triggering separation of the core and latch domains, exposure of the alpha6 dimerization interface, and assembly of BH3-in-groove homodimers that oligomerize into lipidic pores releasing cytochrome c and SMAC. Because Bax provides near-complete functional overlap, single Bak1 nulls are viable and largely unremarkable, and only Bax Bak1 double deficiency reveals the full phenotype, including persistent interdigital webbing, imperforate vaginal closure, accumulated neurons, and frequent perinatal death. Design accordingly: conditional Bak1 flox alleles combined with Bax flox and a tissue-restricted Cre for apoptosis-resistance studies, or groove mutants when distinguishing activation from effector pore formation during BH3 mimetic pharmacology.
