Chrd, the mouse ortholog of human CHRD, encodes chordin, a secreted protein built from four cysteine-rich CR domains that binds BMP2 and BMP4 with high affinity and blocks receptor engagement, establishing the extracellular gradient that patterns dorsoventral axis, pharyngeal apparatus, and inner ear. Its inhibitory activity is itself regulated proteolytically: BMP1 and the tolloid proteases TLL1 and TLL2 cleave chordin to release active ligand, while twisted gastrulation encoded by Twsg1 forms ternary complexes that can either strengthen or relieve inhibition depending on cleavage state. Nulls show strain-dependent pharyngeal, cardiovascular, and ear defects resembling DiGeorge phenotypes with variable perinatal lethality, so background control is as consequential as allele design.
