Human COX15 mutations cause fatal infantile hypertrophic cardiomyopathy and Leigh syndrome, phenotypes making cardiac and neuronal conditional alleles the most informative designs, while hypomorphic knockins model residual heme A synthesis. Cox10 works in the same pathway and cannot substitute for the synthase step. Use Myh6-MerCreMer or Nes-Cre with echocardiography, COX and SDH histochemistry, complex IV spectrophotometry, and blue native assembly profiling matched to the question.
