Why model Cox15

Cox15 encodes the mouse ortholog of human COX15, the inner membrane heme A synthase that completes cofactor maturation by hydroxylating and oxidizing a methyl group of heme O to yield heme A, the reaction immediately downstream of the farnesylation catalyzed by Cox10. Heme A is required for MT-CO1 maturation and for the heme a and heme a3 centers that drive electron transfer to oxygen, so blocking this step produces isolated cytochrome c oxidase deficiency with degradation of unassembled subunit 1. Human COX15 mutations cause fatal infantile hypertrophic cardiomyopathy and Leigh syndrome, phenotypes making cardiac and neuronal conditional alleles the most informative designs, while hypomorphic knockins model residual heme A synthesis. Cox10 works in the same pathway and cannot substitute for the synthase step. Use Myh6-MerCreMer or Nes-Cre with echocardiography, COX and SDH histochemistry, complex IV spectrophotometry, and blue native assembly profiling matched to the question.
