Cox7a1 Knockout mouse models

Cox7a1, ortholog of human COX7A1, encodes the heart and skeletal muscle isoform of cytochrome c oxidase subunit 7A, a small transmembrane subunit positioned near the dimer interface and the cardiolipin binding region, where it stabilizes the holoenzyme and influences supramolecular organization within respiratory supercomplexes. Deletion in mouse is viable but produces dilated cardiomyopathy with reduced cytochrome c oxidase activity, an established phenotype making this locus useful for studying isoform specific contributions to cardiac bioenergetics rather than for modeling absolute complex IV loss. Consequently, a conditional null resolves whether the cardiomyopathy is cardiomyocyte autonomous or secondary, while a knockin swapping in the ubiquitous isoform sequence tests isoform specificity directly. The paralog Cox7a2 is co-expressed and partially compensates, which explains the graded phenotype and argues for compound alleles when full loss is required. Pair Myh6-MerCreMer or Ckmm-Cre with echocardiography, respirometry, supercomplex blue native analysis, and exercise testing.

Whole body loss of Cox7a1 gives the clearest readout when the question is whether the gene is required at all. Follow up tissue work can still move to a conditional allele if lethality or compensation appears.
