No paralog compensates, since CD28 is the functional antagonist rather than a substitute. Humanized CTLA4 knockins are essential for ipilimumab and Fc-engineered antibody work, and haploinsufficient alleles suit human CTLA4 insufficiency and LRBA deficiency biology.
