Ctsg, ortholog of human CTSG, encodes cathepsin G, a chymotrypsin-family serine protease stored in mature form within neutrophil azurophilic granules alongside elastase and proteinase 3. Cathepsin G is unusual in possessing dual chymase-like and tryptase-like specificity, allowing it to process angiotensin, activate PAR4 on platelets, convert chemokines and cytokines, kill bacteria through combined catalytic and cationic antimicrobial activity, and decorate neutrophil extracellular traps. Maturation of cathepsin G, elastase, and proteinase 3 all requires dipeptidyl peptidase I (cathepsin C, Ctsc) removal of the activation dipeptide, so Ctsc deficiency phenocopies triple protease loss and offers a useful genetic shortcut. Because Elane and Prtn3 overlap substantially in substrate range and granule localization, single Ctsg nulls show only partial defects in bacterial killing and compound alleles are needed for clear phenotypes. Reserve humanization for inhibitor pharmacology where chymase pocket specificity differs by species, and use granulocyte-restricted conditional deletion for tissue injury endpoints.
