The proenzyme is trafficked by mannose-6-phosphate receptors and activated at acidic pH by autocatalysis or by cathepsin D and legumain, and its substrates span bulk lysosomal turnover, maturation of other proenzymes, extracellular matrix components after secretion, and inflammatory mediators, with lysosomal leakage of cathepsin B implicated in NLRP3 inflammasome activation, tumor invasion, and necrotic death. Nulls are viable and fertile, partly because cathepsins L, Z, and S share substrate space, so meaningful phenotypes often require compound Ctsb Ctsl or Ctsb Ctsz alleles. Use conditional deletion with myeloid or tumor cell Cre when localizing the relevant protease pool, and active site or occluding loop knockins when endopeptidase versus exopeptidase activity is the mechanistic question.
