Why model Dlat
Dlat is the mouse ortholog of human DLAT and encodes the E2 dihydrolipoamide S-acetyltransferase that forms the trimeric 60-subunit core of the pyruvate dehydrogenase complex, presenting lipoyl domains that swing acetyl groups from E1 to coenzyme A and thereby committing glycolytic pyruvate to acetyl-CoA and the TCA cycle. The lipoyl arm, installed by LIAS and LIPT1, also makes E2 the docking platform for E1, E3 and E3BP, so its loss removes the scaffold on which flux control by Pdk kinases and Pdp phosphatases operates. Complete deficiency produces congenital lactic acidosis, so conditional nulls with Alb-Cre, Ckmm-Cre or Nes-Cre are the practical route for isolating tissue-specific pyruvate oxidation defects, while patient missense knockins retain partial activity and model graded disease. Lipoylated E2 is also the dominant autoantigen of primary biliary cholangitis, making humanized alleles attractive for immunological endpoints; match allele to readout, whether carbon 13 pyruvate tracing, blood lactate, or PDH activity assays.
