Mouse Dld, ortholog of human DLD, specifies the FAD-dependent homodimeric E3 dihydrolipoamide dehydrogenase shared by four mitochondrial systems, pyruvate dehydrogenase, alpha-ketoglutarate dehydrogenase, branched-chain ketoacid dehydrogenase and the glycine cleavage complex, where it reoxidizes reduced lipoamide and passes electrons to NAD+. This shared architecture explains why human DLD mutations cause combined E3 deficiency with simultaneous lactic acidemia, elevated alpha-ketoglutarate, branched-chain ketoaciduria and sometimes hyperglycinemia, and why the enzyme is itself a significant source of reactive oxygen species. Because a global null removes four dehydrogenase activities at once and is not survivable, conditional alleles are the workable design, with hepatocyte, neuronal or cardiomyocyte Cre lines separating hepatic decompensation from encephalopathy and cardiomyopathy.
