Eif4ebp1, mouse ortholog of human EIF4EBP1, encodes 4E-BP1, the principal translational repressor downstream of mTORC1, which binds eIF4E through a canonical YXXXXL phi motif plus a secondary elongated interface and thereby blocks eIF4G recruitment and cap-dependent initiation. RAPTOR docks the TOS motif and mTORC1 phosphorylates a hierarchical series beginning at threonines 37 and 46, then threonine 70 and serine 65, progressively releasing eIF4E and licensing translation of 5 prime TOP and eIF4E-sensitive mRNAs including cyclin D1, MCL1 and MYC. Because 4E-BP1 phosphorylation is rapamycin-resistant relative to S6K1, it is the readout that distinguishes true mTOR kinase inhibitors from rapalogs. Nonphosphorylatable knockin alleles carrying alanine substitutions at the four regulatory sites give constitutive repression and are far more informative than a null for testing whether a tumor depends on cap-dependent translation. Eif4ebp2 and Eif4ebp3 compensate, especially in brain, so compound alleles are needed before concluding requirement.
