Why model F9

F9, ortholog of human F9, encodes coagulation factor IX and sits on the X chromosome, making hemizygous males the natural genetic configuration for hemophilia B modeling. The vitamin K dependent zymogen presents a Gla domain, two EGF modules, an activation peptide, and a protease domain; cleavage by factor XIa or by the tissue factor and factor VIIa complex yields factor IXa, which assembles with factor VIIIa on activated platelet membranes into intrinsic tenase to activate factor X, with the EGF1 and Gla surfaces mediating cofactor and membrane contacts. Mouse nulls reproduce the coagulopathy and remain the standard preclinical hemophilia B platform. Two allele designs answer different questions: patient-specific missense knockins interrogate residual activity, inhibitor development, and immune tolerance, whereas the hyperfunctional Padua substitution at Arg338 underpins gene therapy potency studies. Humanized F9 knockins are required when human factor IX directed antibody, Fc or albumin fusion pharmacokinetics, or mimetic bispecific activity is the readout.
