Why model Fancd2

Fancd2, the mouse ortholog of human FANCD2, encodes the central effector of the Fanconi anemia pathway, forming an obligate heterodimer with FANCI that clamps around double stranded DNA at stalled forks and is monoubiquitinated at lysine 561 by the FANCL RING subunit of the core complex, a modification that locks the ID2 clamp on chromatin and recruits SLX4 with the XPF and MUS81 nucleases to unhook interstrand crosslinks, after which USP1 and UAF1 remove the mark to allow release. FANCD2 also protects nascent strands at stalled forks from MRE11 degradation independently of crosslink unhooking. That functional split dictates allele design: a non-ubiquitinatable K559R knockin isolates the ubiquitin-dependent repair arm while preserving protein, whereas a null removes both activities, and USP1 alleles test the release step. Null mice show crosslinker sensitivity, germ cell loss and epithelial cancers, are viable, and become marrow failure models on aldehyde-sensitized backgrounds. Choose the knockin when mechanism, not dependency, is the question.
