Why model Fanca
Fanca, mouse ortholog of human FANCA, encodes the most frequently mutated Fanconi anemia gene product, a scaffolding subunit of the multiprotein FA core complex that, with FANCB, FANCC, FANCE, FANCF, FANCG, FANCL and FAAP proteins, functions as the E3 ubiquitin ligase that monoubiquitinates FANCD2 and FANCI after the FANCM and FAAP24 sensor detects a stalled replication fork at an interstrand crosslink. Loss abolishes ID2 activation, blocking recruitment of SLX4, XPF and the translesion machinery, producing the diagnostic hypersensitivity to mitomycin C and diepoxybutane and progressive bone marrow failure. Null mice reproduce the cellular crosslinker sensitivity and germ cell attrition but only develop overt marrow failure when combined with aldehyde detoxification alleles such as Aldh2 or Adh5, which is the key design insight for building a faithful hematopoietic model. Because the core complex is nonredundant, single nulls are informative. Use constitutive nulls for crosslink repair biology and compound aldehyde-stress backgrounds for marrow failure endpoints.
