Fcer1g, the mouse ortholog of human FCER1G, encodes the FcR gamma chain, a compact ITAM-bearing transmembrane subunit that supplies signaling competence to ligand-binding alpha chains including FcgammaRI, FcgammaRIII, FcgammaRIV, FcepsilonRI, and several PIR and OSCAR family receptors. Receptor crosslinking by immune complexes drives Src family phosphorylation of the ITAM tyrosines, Syk recruitment through tandem SH2 domains, and downstream PLCgamma2, PI3K, and Vav signaling that produces degranulation, phagocytosis, and antibody dependent cytotoxicity. Because the gamma chain is also required for surface assembly and stability of its partner alpha chains, a germline null collapses essentially all activating Fc receptor output at once, which is powerful for asking whether a phenotype is Fc dependent but useless for resolving which individual receptor carries it. Conditional deletion in myeloid or NK compartments, or reconstitution on a null background with single humanized alpha chains, keeps interpretation clean, and note that Tyrobp supplies parallel ITAM coupling for a distinct receptor set.
