Fcer1a encodes the alpha chain of the high-affinity IgE receptor, mouse ortholog of human FCER1A, providing the ligand-binding subunit of FcepsilonRI, a tetramer completed by the beta chain MS4A2 and a disulfide-linked FCER1G gamma dimer on mast cells and basophils. Two extracellular Ig-like domains grip the Cepsilon2 to Cepsilon3 region of IgE Fc with picomolar affinity and slow dissociation, so receptors remain chronically occupied; antigen-driven crosslinking of bound IgE aggregates receptors, triggering LYN and FYN phosphorylation of the gamma chain ITAMs, SYK recruitment, LAT and PLCgamma signaling, calcium flux, degranulation of histamine and proteases, eicosanoid generation, and cytokine transcription. Species specificity is the dominant design constraint, since human IgE does not bind mouse FcepsilonRI alpha, which is why humanized FCER1A knockins are required for omalizumab, ligelizumab, and human IgE dependent anaphylaxis studies, whereas conditional deletion with mast cell restricted Cre resolves which effector population drives an allergic endpoint.
