Why model Gba
Gba, the mouse ortholog of human GBA1, encodes lysosomal acid beta-glucocerebrosidase, a retaining glycosidase that cleaves glucosylceramide into ceramide and glucose, depends on saposin C for activity, and reaches the lysosome via LIMP2 (Scarb2) rather than the mannose-6-phosphate route. Biallelic loss causes Gaucher disease with glucosylsphingosine accumulation, whereas single variant alleles such as N370S and L444P are the strongest common genetic risk factors for Parkinson disease, acting through impaired lysosomal flux and a reciprocal loop between reduced enzyme activity and alpha-synuclein aggregation. Constitutive nulls die within a day of birth from a defective epidermal permeability barrier, so conditional deletion with nestin-Cre or microglial drivers is required for adult neuropathology, while point knockins preserve residual activity and model heterozygous risk. The non-lysosomal paralog Gba2 hydrolyzes the same substrate elsewhere, so interpret lipid panels accordingly and match allele class to storage, chaperone response, or synucleinopathy endpoints.
