Gckr, the mouse ortholog of human GCKR, encodes glucokinase regulatory protein, a sugar isomerase fold protein that binds hepatic glucokinase and sequesters it in the nucleus, releasing it to the cytosol when fructose-1-phosphate rises and retaining it when fructose-6-phosphate accumulates, so hepatic glucose phosphorylation reports fructose and glycolytic intermediates rather than glucose alone. The common human P446L variant weakens this restraint, lowering fasting glucose and insulin while raising triglycerides, one of the clearest examples of an allele with divergent cardiometabolic consequences. That biology argues for a humanized knockin carrying the P446L substitution when the goal is to reproduce the human tradeoff, whereas a hepatocyte conditional null models complete loss of glucokinase sequestration and unrestrained lipogenic flux. No paralog substitutes for GKRP, so residual regulation depends on glucokinase expression itself.
