Why model Grem1
Grem1 is the mouse ortholog of human GREM1 and encodes gremlin 1, a secreted DAN-family cysteine-knot protein that dimerizes and clamps BMP2, BMP4, and BMP7 to prevent type I and type II receptor engagement, thereby sustaining the BMP, SHH, and FGF feedback loop that maintains limb bud outgrowth, with additional reported engagement of VEGFR2 independent of BMP. Nulls die neonatally with failed lung septation and renal agenesis, making conditional deletion the only practical route to adult fibrosis and tumor stroma questions. The human disease logic is ectopic expression rather than loss, since duplication of an upstream enhancer region drives GREM1 in intestinal crypt epithelium and causes hereditary mixed polyposis, so an enhancer duplication or epithelium-restricted overexpression knockin models the disease far better than a null.
