Grem1 is the mouse ortholog of human GREM1 and encodes gremlin 1, a secreted DAN-family cysteine-knot protein that dimerizes and clamps BMP2, BMP4, and BMP7 to prevent type I and type II receptor engagement, thereby sustaining the BMP, SHH, and FGF feedback loop that maintains limb bud outgrowth, with additional reported engagement of VEGFR2 independent of BMP.
