Why model Hey1
Belonging to the HESR family, Hey1 is the mouse ortholog of human HEY1 and encodes a basic helix-loop-helix Orange domain repressor that is a direct Rbpj-dependent Notch target; it homodimerizes and heterodimerizes with Hey2 and Hes1, binds class B and class C sites, and represses cardiogenic and chondrogenic programs including Gata4-dependent transcription, contributing to arterial versus venous endothelial specification and cardiac cushion development. In humans, the recurrent HEY1 to NCOA2 fusion defines mesenchymal chondrosarcoma, which makes a fusion knockin a distinct and highly informative allele class from simple loss: the fusion asks whether a chimeric transcriptional activator initiates cartilage-lineage tumors, whereas a conditional null asks whether endogenous repression is required for vascular and valvular patterning. Single Hey1 nulls are viable because Hey2 and Hes1 compensate, and double Hey1 and Hey2 animals show the cardiovascular lethality, so plan compound genetics and an endothelial or mesenchymal driver alongside the primary allele.
