Somatic G12V and Q61L substitutions occur in bladder and head and neck carcinoma, while germline G12S and G13D cause Costello syndrome, giving three distinct allele classes: a Lox-Stop-Lox oncogenic knockin for tumor initiation, an endogenous RASopathy point mutation for developmental and cardiac phenotypes, and humanization for isoform-selective inhibitor testing. Hras nulls are viable because Kras carries the essential load, so single-gene deletion is rarely informative, and compound Hras and Nras alleles combined with tissue-restricted Cre give interpretable results.
