Ifna1 sits within a tandem cluster of roughly fourteen intact interferon alpha genes on mouse chromosome 4 and corresponds to human IFNA1, encoding a single helical type I interferon that engages the shared IFNAR1 and IFNAR2 heterodimer, activates JAK1 and TYK2, and assembles the ISGF3 complex of STAT1, STAT2, and IRF9 on interferon stimulated response elements. Subtype identity tunes receptor affinity and therefore the balance of antiviral versus antiproliferative output rather than creating unique signaling, which has a hard consequence for allele design: deleting Ifna1 alone is almost always uninformative because the remaining subtypes are coregulated by IRF7 and compensate fully. Productive designs instead remove the entire cluster, target Ifnar1, or install a reporter at Ifna1 to time and localize subtype induction in vivo. When human interferon pharmacology is the endpoint, species restricted receptor engagement makes humanization of the ligand or receptor pair, not a subtype knockout, the appropriate allele.
