Conditional alleles with nephron segment-restricted Cre drivers separate thick ascending limb from collecting duct contributions, while knockins of specific pH-sensing or PIP2-binding residues test gating mechanism, and humanized pore alleles support ROMK inhibitor diuretic programs.
