Kcnj16, the mouse ortholog of human KCNJ16, encodes Kir5.1, a subunit that is electrically silent on its own and functions only by heteromerizing with Kir4.1 or Kir4.2, where it converts a weakly pH-sensitive channel into a steeply pH-sensitive one through a lysine in the second transmembrane domain of its partner and through a Kir5.1-specific carboxy-terminal interface. Kir4.1/Kir5.1 channels operate on the basolateral membrane of the distal convoluted tubule and collecting duct, in astrocytes, in carotid body and brainstem chemosensitive neurons, and in retina, making the subunit a molecular link between carbon dioxide, intracellular pH, and potassium conductance. Null rats and mice show impaired potassium and acid-base handling with hypokalemic alkalosis and altered chemoreflex responses. Because the obligate partnership is with Kcnj10 and Kcnj15 products, deletion of Kcnj16 does not abolish current but shifts pH sensitivity, so design endpoints around gating and transport physiology rather than total conductance, using nephron or glia-restricted Cre.
